  • Results: We found that the Sig-1R is a new molecular chaperone that shares no homology with any mammalian proteins. The novel characteristics of the Sig-1R chaperone include:(1) Sig-1Rs localize specifically at the mitochondrionassociated ER membrane (MAM) and regulate direct Ca2+ influx from ER to mitochondria by stabilizing inositol 1, 4, 5- trisphosphate receptors (IP3Rs) at the MAM;(2) Synthetic compounds possessing affinities for Sig-1Rs can influence the Sig-1R chaperone activity by regulating the association between Sig-1Rs and another ER chaperone BiP. Conclusion: Mitochondrial Ca2+ supplied by IP3Rs results in the activation of key enzymes in the TCA cycle. Thus, Sig-1Rs regulating Ca2+ signaling between ER and mitochondria may play important roles in bioenergetics and cell survival. Molecular chaperones that combat against the formation of protein …
